Differential Cell Counter

A differential cell counter is a laboratory instrument or method that measures the number and relative proportions of distinct cell types within a mixed sample. As cells pass individually through a sensing region, their size, internal complexity, light-scattering properties, fluorescence, or electrical impedance generate signals that enable classification and counting. In genetics, differential cell counting helps characterize cellular phenotypes associated with gene variants, compare engineered and control populations, and assess changes during cell culture or experimental treatment. Reliable population measurements support studies of gene function, cellular heterogeneity, disease mechanisms, and the reproducibility of molecular and cellular assays.

The Moxi Z miniature automated cell counter is a novel instrument that combines the Coulter Principle with patented thin-film sensor technology and a proprietary software algorithm to perform sizing and counting of a broad size range of particles as well as to determine the overall health of monodisperse mammalian cell cultures. This protocol describes the use of this instrument for counting and assessing the health of cell cultures.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.
