Eosinophil Stat3

Eosinophil STAT3 refers to the STAT3 transcription factor and its role in regulating gene expression in eosinophils, immune cells involved in host defense and allergic inflammation. Cytokine binding to cell-surface receptors activates associated Janus kinases, which phosphorylate STAT3; STAT3 then dimerizes, enters the nucleus, and binds regulatory DNA sequences to alter transcription. This signaling pathway can influence eosinophil development, survival, activation, and inflammatory mediator production. Studying eosinophil STAT3 in genetics helps clarify how regulatory variants or altered signaling contribute to asthma, allergies, and other eosinophil-associated disorders, while supporting the identification of potential molecular targets.

Tandem splicing events occur at sites less than 12 nucleotides apart. Quantifying ratios of such splice variants is feasible using an absolute quantitative PCR approach. This manuscript describes how splice variants of the gene STAT3, in which two splicing events results in Serine-701 inclusion/exclusion and α/β C-termini, can be quantified.

2020

Environmental allergens such as house dust mites (HDM) often contain microbial substances that activate innate immune responses to regulate allergic inflammation. The protocol presented here demonstrates the identification of dsRNA species in HDM allergens and characterization of their immunogenic activities in modulating eosinophilic lung inflammation.
