Functional Characterization

Functional characterization is the systematic investigation of how a gene, genetic variant, or regulatory element influences biological function and phenotype. In genetics, researchers combine approaches such as gene expression analysis, targeted mutation, loss- or gain-of-function experiments, and protein or pathway assays to connect a DNA sequence with its molecular and cellular effects. These studies help distinguish disease-causing variants from benign changes, clarify gene pathways, and validate candidate genes identified through sequencing or association studies. Functional characterization therefore supports disease mechanism research, diagnostic interpretation, therapeutic target discovery, and the development of more accurate genetic models.

Current knowledge on the cellular basis of cardiac diseases mostly relies on studies on animal models. Here we describe and validate a novel method to obtain single viable cardiomyocytes from small surgical samples of human ventricular myocardium. Human ventricular myocytes can be used for electrophysiological studies and drug testing.
