Polyacrylamide Embedding

Polyacrylamide embedding is a laboratory technique that immobilizes biological specimens within a porous polyacrylamide matrix, creating a stable support for microscopy and molecular analysis. The gel forms when acrylamide and a cross-linker polymerize, typically through an initiator and accelerator system; adjusting the monomer-to-cross-linker ratio changes pore size, stiffness, and sample accessibility. In genetics, this approach can help preserve specimen structure during processing and support the detection or spatial analysis of nucleic acids, proteins, or cellular features. By combining physical stabilization with tunable material properties, polyacrylamide embedding improves handling and reproducibility in imaging-based genetic research.

2026

Although extracellular matrix (ECM) stiffness is a key regulator of cellular behavior, the underlying sensing and transduction mechanisms remain unclear. To address this, we provide protocols for creating polyacrylamide- and silicone-based substrates with tunable stiffness to advance our understanding of cell-ECM crosstalk.
