Fungal Dna Isolation

Fungal DNA isolation is a laboratory technique used to extract genomic DNA from fungal cells for genetic analysis. Because fungal cells have resilient walls, the process typically combines mechanical or chemical disruption with cell lysis, followed by removal of proteins, polysaccharides, and other contaminants and purification of the DNA. The resulting nucleic acid provides a template for polymerase chain reaction, genotyping, sequencing, and other molecular assays. Reliable isolation supports studies of fungal diversity, inheritance, evolution, pathogenicity, and gene function, while DNA quality and purity strongly influence the accuracy and reproducibility of downstream genetic analyses.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.
