L1 Animal Seeding

L1 animal seeding is a laboratory method for placing first-stage larvae, typically of C. elegans, onto a defined culture surface. It enables researchers to begin experiments with animals at a comparable developmental stage: newly hatched larvae are collected and distributed at a controlled density onto nematode growth medium plates containing food or an experimental treatment. This synchronization reduces variation caused by age differences and supports reproducible analysis of development, phenotype, gene function, and environmental responses. In genetics, L1 seeding helps compare mutant and wild-type strains under consistent conditions, improving the interpretation of developmental assays and screening experiments.

A method for seeding titanium blood-contacting biomaterials with autologous cells and testing biocompatibility is described. This method uses endothelial progenitor cells and titanium tubes, seeded within minutes of surgical implantation into porcine venae cavae. This technique is adaptable to many other implantable biomedical devices.

We present the procedure to assemble a seed coat bedding assay (SCBA) from Arabidopsis thaliana seeds. The SCBA was shown to be a powerful tool to explore genetically and in vitro how the endosperm controls seed germination in dormant seeds and in response to light cues. The SCBA is in principle applicable under any situation where the endosperm is suspected to influence embryonic growth.
