Oogenesis Analysis

Oogenesis analysis is the study of how female germ cells develop into mature oocytes, providing insight into inheritance, fertility, and reproductive health. It examines processes including mitotic proliferation, meiotic recombination, folliculogenesis, and the unequal divisions that produce one large egg and polar bodies. In genetics, this analysis helps researchers evaluate chromosome segregation, genomic imprinting, and sources of aneuploidy that can affect embryo viability. It also supports investigations of infertility, maternal-age effects, reproductive disorders, and developmental abnormalities, while advancing understanding of how genetic variation is generated and transmitted across generations.

In human women, oogenesis produces one mature egg cell or ovum for every precursor cell that enters meiosis. This process differs in two unique ways from the equivalent procedure of spermatogenesis in males. First, meiotic divisions during oogenesis are asymmetric, meaning that a large oocyte (containing most of the cytoplasm) and minor polar body are produced as a result of meiosis I, and again following meiosis II. Since only oocytes will go on to form embryos if fertilized, this unequal...

Oogenesis, the process of developing egg cells (female gametes), occurs within the ovaries and is fundamental to female fertility. This sequence begins during fetal development when diploid oogonia in the developing ovaries undergo mitotic divisions to produce primary oocytes. By birth, these primary oocytes enter prophase I of meiosis but become arrested in this stage, remaining suspended until puberty. Each primary oocyte is surrounded by a layer of pre-granulosa cells, forming what is known...

2022

The basement membrane is essential for tissue and organ morphogenesis during development. To better understand the mechanisms leading to proper placement of this structure, the protocol presented describes methods to visualize and characterize the intracellular trafficking and secretion of basement membrane proteins in epithelial cells using confocal and super-resolution microscopy.

Automated Analysis of Intracellular Phenotypes of Salmonella Using ImageJ

Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.
