Syncytial Stage

The syncytial stage is a developmental phase in which multiple nuclei share a common cytoplasm because nuclear divisions occur without cytokinesis, making it a key model for understanding early genetic patterning. In the Drosophila embryo, rapid, synchronized mitoses produce nuclei that migrate toward the cortex, where maternal RNA and protein gradients regulate gene expression before cellularization partitions the embryo into individual cells. Studying this stage reveals how maternal-effect genes, morphogens, and nuclear interactions establish body axes and activate the zygotic genome. It also provides a framework for investigating cell-cycle control, spatial gene regulation, and the transition from a shared cytoplasm to cellular organization.

Source: Bouillier, C., et al. Generation, Amplification, and Titration of Recombinant Respiratory Syncytial Viruses. J. Vis. Exp. (2019)The video demonstrates a plaque titration assay used to quantify infectious recombinant respiratory syncytial virus (RSV) in epithelial cell monolayers. It presents the infection of host cells with serially diluted viral suspensions, the restriction of viral spread by a cellulose-based semi-solid overlay, and the formation of localized plaques from cytopathic...

Respiratory Syncytial Virus Disease

2026

Human respiratory syncytial virus (RSV) is a widespread pathogen that primarily targets infants and young children but also poses a serious health risk to elderly and immunocompromised individuals. Belonging to the Pneumoviridae family, RSV is a negative-sense, single-stranded RNA virus within the Pneumovirus genus. Its global health burden is significant, with millions of cases annually resulting in hospitalizations and mortality, particularly in resource-limited settings. Although most...

For in-depth mechanistic analysis of the respiratory syncytial virus (RSV) RNA synthesis, we report a protocol of utilizing the chaperone phosphoprotein (P) for coexpression of the RNA-free nucleoprotein (N0) for subsequent in vitro assembly of the virus-specific nucleocapsids (NCs).

An Assay to Measure the Neutralizing Antibody Titer Specific for Respiratory Syncytial Virus

This video demonstrates an assay for measuring neutralizing antibodies against respiratory syncytial virus (RSV). Pre-incubation with serum containing RSV-neutralizing antibodies neutralizes the virus particles, reducing cellular infection and plaque formation. Virus-infected cells are then subjected to fixation, blocking, and fluorophore-conjugated antibody labeling for plaque visualization. This process enables the identification of the highest serum dilution that neutralizes fifty percent of...
