In Vitro Suppression Assay

An in vitro suppression assay is an experimental method used to measure how effectively one immune cell population inhibits the activation or function of another under controlled laboratory conditions. Typically, responder T cells are stimulated through antigen receptors or mitogenic signals and cultured alone or with potential suppressor cells, such as regulatory T cells; suppression is assessed by comparing responder-cell proliferation, cytokine production, or activation markers. In immunology and infection research, the assay helps characterize regulatory mechanisms, evaluate immune tolerance, and determine how pathogens or candidate therapies alter cellular immune responses. Results can clarify interactions that shape inflammation, antimicrobial defense, and disease progression.

Human In Vitro Suppression as Screening Tool for the Recognition of an Early State of Immune Imbalance

2011

Tregs are potent suppressors of the immune system. There is a lack of unique surface markers to define them, hence, definitions of Tregs are primarily functional. Here we describe an optimized in vitro assay capable of identifying immune imbalance in subjects at risk to develop T1D.

Co-Infiltration Assay for Screening Pathogen Effectors that Suppress Plant RNA Silencing

Source: Shi, J., et al. Screening and Identification of RNA Silencing Suppressors from Secreted Effectors of Plant Pathogens. J. Vis. Exp. (2020)This video demonstrates a co-infiltration assay for identifying pathogen effector proteins that suppress RNA silencing in Nicotiana benthamiana 16c. A fluorescent reporter and a candidate effector gene are delivered into plant leaves via Agrobacterium. Normally, RNA silencing degrades reporter transcripts, reducing fluorescence. Suppressor effectors...

2012

Amblyopia is a developmental disorder of the visual cortex that is often accompanied by strong suppression of one eye. We present a new technique for measuring and treating interocular suppression in patients with amblyopia that can be deployed using virtual reality goggles or a portable iPod Touch device.

Commonly used, highly accessible methods for examining cell migration and invasion in vitro are described. The first method is the cell wound closure assay that measures cell motility. The second method is the transwell migration and invasion assay that assesses the chemotactic and invasive capacity of cells.
