Bone Graft Alternatives

Bone graft alternatives are biomaterials or biologic strategies used to support bone repair when an autograft or donor bone is unavailable, insufficient, or undesirable. Their mechanisms vary: porous scaffolds can provide an osteoconductive framework for cell attachment and tissue ingrowth, while selected biologic materials may supply signals that promote osteogenic activity; gradual resorption can allow replacement by new bone. In medicine, these alternatives are applied in fracture healing, spinal fusion, dental reconstruction, and orthopedic implant procedures. Their use may reduce donor-site complications and expand treatment options, although performance depends on material properties, defect conditions, vascularization, and the patient’s capacity for bone regeneration.

Bone metastasis models do not develop metastasis uniformly or with a 100% incidence. Direct intra-osseous tumor cell injection can result in embolization of the lung. We present our technique modeling primary bone tumors and bone metastasis using solid tumor graft implantation into bone, leading to reproducible engraftment and growth.

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.

Implantation of Inferior Vena Cava Interposition Graft in Mouse Model

To improve our knowledge of cellular and molecular neotissue formation, a murine model of the TEVG was recently developed. The grafts were implanted as infrarenal vena cava interposition grafts in C57BL/6 mice. This model achieves similar results to those achieved in our clinical investigation, but over a far shortened time-course.

In this video, we describe a method for the detection of osteoblast-secreted calcium deposits using an anionic dye, Alizarin Red S. The cultured osteoblasts are first stained and then the dye is recovered for colorimetric quantification of calcium deposits or the degree of osteoblast mineralization.
