Bone Volume Analysis

Bone volume analysis is the quantitative assessment of the amount and spatial distribution of bone within an anatomical region or tissue sample, supporting evaluation of skeletal structure and health. It typically uses medical imaging or three-dimensional microscopy to segment bone from surrounding tissue, reconstruct its geometry, and calculate measures such as total volume, bone volume fraction, and regional changes. In medicine, these measurements help characterize osteoporosis, fracture healing, implant integration, and skeletal development. Standardized analysis can also improve comparisons across patients, experimental models, and treatment groups, providing objective evidence for diagnosis, monitoring, and research into bone disease and regeneration.

Undecalcified bone histology provides important information for a variety of clinical and research applications. It is technically challenging, particularly with large size specimens. This video illustrates the process of producing good quality sections and demonstrates the technical difficulties and methods with which to overcome them.

Segmentation of three-dimensional data from many imaging techniques is a major bottleneck in analysis of complex biological systems. Here, we describe the use of SuRVoS Workbench to semi-automatically segment volumetric data at various length-scales using example datasets from cryo-electron tomography, cryo soft X-ray tomography, and phase contrast X-ray tomography techniques.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
