Chemoresistance

Chemoresistance is the ability of cancer cells or other disease-causing cells to survive treatment with drugs that would normally inhibit or destroy them, limiting therapeutic success in medicine. It can arise through reduced drug uptake, increased drug efflux, enzymatic drug inactivation, enhanced DNA repair, altered drug targets, or resistance to programmed cell death. Chemoresistance may exist before treatment or develop as cells adapt under drug pressure, and resistant populations can drive disease relapse and progression. Studying these mechanisms supports biomarker development, treatment selection, combination therapies, and the design of new drugs that overcome resistance.

Cancer stem cells (CSCs) are implicated in tumor relapse due to chemoresistance. We have optimized a protocol for selection and expansion of CSCs from ovarian cancer cell lines. By treating cells with the chemotherapeutic cisplatin and culturing in a stem cell promoting media we enrich for non-adherent CSC cultures.
