Gene Disease Triads

Gene-disease triads are structured relationships that connect a gene, a disease-associated variant, and the clinical phenotype it can produce, helping researchers interpret genetic causes of illness. A pathogenic variant may alter protein sequence, gene expression, or dosage, disrupting a cellular pathway and generating characteristic signs and symptoms; confidence in the relationship increases when inheritance patterns, patient phenotypes, population frequency, and functional evidence agree. In medicine, triad-based analysis supports variant interpretation, diagnosis of rare disorders, genetic counseling, and selection of patients for targeted testing or treatment. It also helps organize evidence for gene discovery and precision medicine.

Lipoxygenase (LOX) isozymes can generate products that may increase or decrease neuroinflammation and neurodegeneration. A gene-environment interaction study could identify LOX isozyme-specific effects. Using the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) model of nigrostriatal damage in two LOX isozyme-deficient transgenic lines allows for comparison of the contribution of LOX isozymes on dopaminergic integrity and inflammation.
