Liver Repopulation Level

Liver repopulation level is a measure of how much of a recipient’s liver tissue has been replaced or functionally occupied by introduced cells, making it an important indicator of regenerative success. It reflects the extent to which transplanted or engineered hepatocytes engraft, survive, proliferate, and contribute to hepatic tissue and function within the host liver. In medicine, researchers use this parameter to evaluate cell-based therapies, compare repopulation strategies, and assess whether damaged liver tissue can regain metabolic capacity. Quantifying it connects cellular engraftment with treatment outcomes and supports the development of regenerative approaches for liver disease.

This protocol describes decellularization of Sprague Dawley rat kidneys by antegrade perfusion of detergents through the vasculature, producing acellular renal extracellular matrices that serve as templates for repopulation with human renal epithelial cells. Recellularization and use of the resazurin perfusion assay to monitor growth is performed within specially-designed perfusion bioreactors.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.

Exp., (2016)This video demonstrates adeno-associated virus-mediated gene delivery in a humanized rat liver scaffold. The vector delivers a transgene encoding a fluorescent reporter and gene-silencing RNA into liver cells via receptor-mediated endocytosis. This model supports efficient transduction in a perfused 3D liver tissue system.
