Monocytic Lineage

The monocytic lineage is a branch of hematopoiesis that produces monocytes and related phagocytic cells, which support innate immune defense and tissue maintenance. In the bone marrow, hematopoietic stem and progenitor cells progressively differentiate through monoblast and promonocyte stages before releasing circulating monocytes; cytokines and growth factors regulate their development, survival, and activation. After entering tissues, monocytes can differentiate into macrophages or certain dendritic cells, where they engulf pathogens, remove damaged material, and coordinate inflammation. Understanding this lineage helps clinicians interpret blood counts, investigate infections and inflammatory disorders, and recognize monocytic leukemias and other diseases involving abnormal myeloid development.

We describe use of ImageStream technology (www.amnis.com), which combines quantitative flow cytometry with simultaneous high-resolution digital imaging, to quantify cellular mechanisms of primary immune cells from well-defined patient cohorts. Our studies provide a blueprint for translational investigations to quantify lineage specific cellular responses in small samples from subject cohorts.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.
