Muscle Preservation

Muscle preservation is the maintenance of skeletal-muscle mass, strength, and function during aging, illness, injury, weight loss, or prolonged inactivity, making it an important goal in medicine. It depends on balancing muscle-protein synthesis with breakdown: resistance exercise and sufficient dietary protein provide mechanical and nutritional signals that support synthesis, while immobility, inflammation, and inadequate energy intake can accelerate atrophy. Clinicians apply muscle-preservation strategies through progressive rehabilitation, nutrition support, and, when appropriate, management of underlying disease to reduce weakness, improve mobility, and support recovery after hospitalization or surgery. These approaches also inform care for sarcopenia and other conditions associated with functional decline.

Innovative Strategies for Organ Preservation in Heart Transplantation: Uniform Cooling Preservation and Ex-situ Normothermic Perfusion

2025

Donor heart preservation is a major determinant of success in heart transplantation. Technological advances have led to optimizing the conditions of preservation, along with a more accurate assessment of the donor heart before transplantation. We herein describe two innovative technologies currently applied for cardiac preservation: uniform cooling preservation and normothermic ex-situ heart perfusion.

Ex Vivo Assessment of Contractility, Fatigability and Alternans in Isolated Skeletal Muscles

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.

Snap Freezing of Muscle Tissue for Sectioning: A Protocol to Obtain Ultrathin Sections of Rapidly Frozen Murine Muscle Tissue

2026

This protocol includes a step-by-step workflow for calcified vascular specimens: tissue handling, decalcification, RNA validation, calcification level detection, and region-of-interest selection strategies on the Nanostring GeoMx Digital Spatial Profiler (DSP). The goal is to present a comprehensive method for preserving vascular tissue morphology and RNA for reliable spatial transcriptomic analysis.
