Tissue Microarray Construction

Tissue microarray construction is a pathology technique that arranges small, representative tissue cores from multiple donor specimens into a single recipient paraffin block, enabling efficient comparison of biological markers. Using a tissue arrayer, investigators select defined regions from formalin-fixed, paraffin-embedded tissue, transfer the cores into a precisely mapped grid, and section the completed block for parallel staining. The resulting array supports immunohistochemistry, in situ hybridization, and other molecular assays across many samples under consistent experimental conditions. In medicine, this approach conserves valuable specimens, improves assay standardization, and accelerates biomarker validation, diagnostic research, and studies of disease progression or treatment response.

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].

Tissue microarrays allows for an efficient method to gain concurrent information from a multitude of tissues. Representative parts of tissues are assembled into a single paraffin block. Sections from the block are used for immunohistochemistry and analysis of protein expression patterns. Digital scanning generates corresponding images for distribution of data.
