Tumor Histopathology

Tumor histopathology is the microscopic examination of tumor tissue to identify abnormal cellular and architectural features, helping determine whether a lesion is benign or malignant. After biopsy or surgical removal, tissue is fixed, sectioned, stained, and evaluated for patterns such as cellular atypia, disorganized growth, invasion, necrosis, and mitotic activity; immunohistochemistry can further identify lineage and tumor-specific markers. In medicine, these findings support diagnosis, tumor classification, grading, and staging, while guiding treatment selection and prognosis. Histopathologic analysis also provides a foundation for correlating tissue morphology with molecular alterations and for improving cancer research and precision oncology.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.

A Novel High-resolution In vivo Imaging Technique to Study the Dynamic Response of Intracranial Structures to Tumor Growth and Therapeutics

We describe a novel in vivo imaging technique that couples fluorescent chimeric mice with intracranial windows and high-resolution 2-photon microscopy. This imaging platform aids studies of dynamic changes in brain tissue and microvasculature, at a single-cell level, following pathological insults and is adaptable to assess intracranial drug delivery and distribution.

This manuscript describes the development of an animal model that allows for the direct testing of the effects of tumor hypoxia on metastasis and the deciphering the mechanisms of its action. Although the experiments described here focus on Ewing sarcoma, a similar approach can be applied to other tumor types.

Tumor Implantation and Injection of a Tumor Vaccine in a Mouse Model

This video demonstrates a cell-based tumor vaccination assay. A mouse, intradermally injected with B16-F10 melanoma cells for tumor formation, is vaccinated with B16-F10 cells secreting Flt3L cytokine near the developed tumor. The secreted Flt3L from the vaccine promotes the maturation and proliferation of dendritic cells that then present tumor antigens to T cells in lymph nodes, triggering a targeted anti-tumor immune response.
