Vascularized Lung Model

A vascularized lung model is an engineered or ex vivo system that recreates key features of lung tissue together with a functional blood vessel network, enabling more realistic study of pulmonary biology and disease. These models typically organize lung cells, extracellular matrix, and endothelial cells into interconnected airways or alveolar structures, while perfusion through the vascular compartment supports nutrient delivery, gas exchange-related functions, and communication between blood vessels and respiratory tissue. In medicine, vascularized lung models help investigate infection, inflammation, fibrosis, cancer, and vascular injury, while providing platforms for drug testing and toxicity assessment. They may also support regenerative medicine and the development of more predictive alternatives to animal models.

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool. Here, we describe the development of a rat EVLP program and refinements that allow for a reproducible model for future expansion.
