3d Image Stack

A 3D image stack is an ordered series of two-dimensional images captured at successive depths and combined to represent the structure of a specimen in three dimensions. In neuroscience, imaging systems collect optical sections through brain tissue, while software aligns the planes and reconstructs them into a volumetric dataset that preserves spatial relationships between cells, processes, and anatomical regions. These stacks support visualization and quantitative analysis of neuronal morphology, tissue architecture, lesion distribution, and connectivity-related features. They also provide a foundation for segmentation, three-dimensional measurements, and comparisons across experimental conditions, strengthening the study of brain organization and disease-related structural changes.

Scalable engineered blood vessels would improve clinical applicability. Using easily sizable 3D-printed guides, rings of vascular smooth muscle were created and stacked into a tubular form, forming a vascular graft. Grafts can be sized to meet the range of human coronary artery dimensions by simply changing the 3D-printed guide size.

A technique for performing quantitative three-dimensional (3D) imaging for a range of fluid flows is presented. Using concepts from the area of Light Field Imaging, we reconstruct 3D volumes from arrays of images. Our 3D results span a broad range including velocity fields and multi-phase bubble size distributions.

We describe a detailed protocol using high-resolution episcopic microscopy to acquire three-dimensional (3D) images of mouse embryos. This improved protocol utilizes a modified tissue preparation method to enhance penetration of the fluorescent dye, thereby permitting morphometric analysis of both small and large-sized specimens.
