Astrocyte Foot Processes

Astrocyte foot processes are specialized expansions of astrocyte membranes that contact blood vessels, synapses, and the brain’s surfaces, helping organize communication between neural tissue and its environment. Their cytoskeletal organization and membrane proteins enable these fine processes to ensheath vascular walls, regulate extracellular ions and neurotransmitters, and support blood-brain barrier function and neurovascular signaling. In neuroscience, studying their morphology, distribution, and molecular composition helps clarify how astrocytes maintain homeostasis, influence synaptic activity, and couple neuronal demand to local blood flow. Changes in foot-process coverage or structure can also provide clues to vascular dysfunction and neurological disease.

Source: Robert E. Sallis, MD. Kaiser Permanente, Fontana, California, USA The foot is a complex structure composed of numerous bones and articulations. It provides flexibility, is the essential contact point needed for ambulation, and is uniquely suited to absorb shock. Because the foot must support the weight of the entire body, it is prone to injury and pain. When examining the foot, it is important to remove shoes and socks on both sides, so that the entire foot can be inspected and compared.

Astrocytes have been recognized to be versatile cells participating in fundamental biological processes that are essential for normal brain development and function, and central nervous system repair. Here we present a rapid procedure to obtain pure mouse astrocyte cultures to study the biology of this major class of central nervous system cells.

Direct Neuronal Reprogramming of Mouse Astrocytes into Neurons

This video showcases the direct reprogramming of astrocytes expressing transforming factors into neurons. Transformation factors trigger a cascade of molecular events facilitating astrocyte-to-neuron conversion. The process is further supported by supplying specific agents and supplements through a neuronal differentiation medium.

(2019)This video demonstrates the visualization of intracellular cargo transport in cultured astrocytes using a fluorescent acidotropic probe that selectively labels acidic endolysosomal vesicles. Time-lapse imaging and Z-stack acquisition with a confocal microscope allow tracking of fluorescently labeled cargo throughout the three-dimensional cell volume.
