Fear Circuit Mapping

Fear circuit mapping is a neuroscience approach that identifies the brain regions and neural pathways that detect threats, generate defensive responses, and regulate fear, helping explain how emotion is organized in the nervous system. Researchers map connections and activity by tracing neural projections and combining electrophysiology, calcium imaging, functional imaging, or optogenetic manipulation as sensory information engages structures such as the amygdala, hippocampus, hypothalamus, periaqueductal gray, and prefrontal cortex. These maps clarify how fear learning and extinction arise, distinguish protective responses from maladaptive anxiety, and support research into stress-related disorders and potential targeted interventions.

Ex Vivo Optogenetic Dissection of Fear Circuits in Brain Slices

Optogenetic approaches are widely used to manipulate neural activity and assess the consequences for brain function. Here, a technique is outlined that upon in vivo expression of the optical activator Channelrhodopsin, allows for ex vivo analysis of synaptic properties of specific long range and local neural connections in fear-related circuits.

This paper introduces an approach of combining laser scanning photostimulation with whole cell recordings in transgenic mice expressing GFP in limited inhibitory neuron populations. The technique allows for extensive mapping and quantitative analysis of local synaptic circuits of specific inhibitory cortical neurons.

Here, we present a protocol to conduct a variety of behavioral tests in cattle that have been designed to evaluate emotions. A battery of behavioral tests (open field test, startle test, bovine zero maze, exit velocity, pen score, and chute score) were conducted to evaluate different components of animal temperament.
