Human Astrocyte Generation

Human astrocyte generation is the production of human astrocytes from pluripotent stem cells or neural progenitor cells for studying the central nervous system. In directed differentiation, defined culture conditions guide progenitors toward an astroglial fate, followed by expansion and maturation into cells that develop characteristic morphology and astrocyte-associated markers. These cells can model neuron–glia interactions, support investigations of neurodevelopment and disease, and provide platforms for drug screening and toxicity testing. Patient-derived induced pluripotent stem cells further enable comparisons between healthy and disease-associated astrocytes, strengthening mechanistic studies of neurological disorders and potentially informing regenerative strategies.

Generating a Primary Culture of Astrocytes from a Rat Pup Brain

2025

The video demonstrates a technique for culturing primary astrocytes from the rat cortex. The cortex is mechanically dissociated to obtain a homogeneous glial cell suspension, which is then plated on polypeptide-coated plates to promote selective proliferation of astrocytes. Subsequently, the astrocytes are enzymatically dissociated from the plates and transferred to fresh media, allowing the culture of pure astrocytes.

An In Vitro Method to Generate Astrocyte Scaffolds within Hydrogel Micro-columns

2025

This video demonstrates a method to generate three-dimensional astrocyte scaffolds within hydrogel micro-columns. The inside of the microcolumns are coated with collagen. Astrocytes are introduced, which adhere to collagen and, with further incubation, self-assemble into a three-dimensional scaffold.

Generating 3D Co-culture Spheres of Astrocytes and Neurons to Induce Synapse Formation

2025

This video demonstrates a procedure for co-culturing the astrocytes and neurons to form 3D spheres. These 3D spheres provide a distinct platform for examining synapse formation and neural network integration, thereby serving as a potential model for research in developmental neuroscience, neurodegenerative diseases, and neuropharmacological interventions.

Generating and Maintaining Astrospheres Using Human Pluripotent Stem Cells

2025

The video demonstrates a method to generate astrospheres from progenitors from human pluripotent stem cells. It involves the growth and proliferation of stem cells with nutrient media containing growth factors and kinase inhibitors. These cells then aggregate and differentiate into astrocytes, forming astrospheres.
