Human Epilepsy Patients

Human epilepsy patients are people living with epilepsy, a neurological disorder characterized by recurrent seizures caused by abnormal, excessive, and synchronized electrical activity in the brain. Seizures arise when neural circuits become temporarily hyperexcitable, producing changes in movement, sensation, awareness, or behavior that vary with the affected brain regions. In neuroscience research, patients may undergo electroencephalography, brain imaging, or intracranial recordings to identify seizure networks and study human brain function. These clinical observations support diagnosis, guide treatment and surgical planning, and provide valuable insights into neural connectivity, cognition, and the mechanisms underlying seizure disorders.

This protocol describes a procedure to track the evolution of mesial network measures in temporal lobe epilepsy (TLE) patients. It is based on the combination of intracranial recordings with a novel numerical technique for data analysis. Specifically, we present a protocol for network analyses of foramen ovale recordings.

Resting-state functional-connectivity MRI has identified abnormalities in patients with a wide range of neuropsychiatric disorders, including epilepsy due to malformations of cortical development. Transcranial Magnetic Stimulation in combination with EEG can demonstrate that patients with epilepsy have cortical hyperexcitability in regions with abnormal connectivity.

Simultaneous Eye Tracking and Single-Neuron Recordings in Human Epilepsy Patients

We describe a method to conduct single-neuron recordings with simultaneous eye tracking in humans. We demonstrate the utility of this method and illustrate how we used this approach to obtain neurons in the human medial temporal lobe that encode targets of a visual search.

High Frequency Oscillations (HFOs) have emerged as presurgical biomarkers for the identification of the epileptogenic zone in pediatric patients with medically refractory epilepsy. A methodology for the noninvasive recording, detection, and localization of HFOs with simultaneous scalp electroencephalography (EEG) and magnetoencephalography (MEG) is presented.
