Mouse Cortical Tissue

Mouse cortical tissue is the neural tissue forming the outer region of the mouse brain, where layered networks of neurons and glial cells support sensory processing, cognition, and coordinated behavior. Its function depends on synaptic communication: electrical activity travels along neuronal membranes, triggers neurotransmitter release at synapses, and is shaped by ion channels, local circuits, and glial regulation. In neuroscience, freshly isolated cortex, organotypic slices, and primary cortical cultures provide tractable models for examining development, neurophysiology, synaptic plasticity, neurodegeneration, and responses to drugs or injury. Findings from these preparations help connect cellular mechanisms with brain function and disease.

Astrocytes have been recognized to be versatile cells participating in fundamental biological processes that are essential for normal brain development and function, and central nervous system repair. Here we present a rapid procedure to obtain pure mouse astrocyte cultures to study the biology of this major class of central nervous system cells.

This video shows a procedure for generating neuronal cultures from late embryo and early postnatal mouse cortex. These cultures can be used for immunocytochemistry, biochemistry, electrophysiology, calcium and sodium imaging and provide a platform to study the neuronal development of transgenic animals that carry a postnatal lethal gene mutation.
