Nerve Fixation Protocol

Nerve fixation protocols are standardized procedures for preserving nerves or neural tissue so their cellular architecture, molecular components, and spatial relationships remain suitable for analysis. Typically, a fixative such as paraformaldehyde stabilizes tissue by forming chemical cross-links between proteins, while controlled perfusion or immersion promotes even penetration and limits autolysis and structural distortion. In neuroscience, careful fixation supports immunohistochemistry, histological staining, fluorescence microscopy, and electron microscopy by maintaining features such as axons, myelin, neuronal somata, and synaptic organization. Fixation conditions, including reagent choice, concentration, temperature, and exposure time, can strongly influence antigen preservation and imaging quality, making protocol selection essential for reliable comparisons across experiments.

We present a protocol for direct vagus nerve injection in rats, enabling drug delivery directly into the nerve without post-injection complications. This method applies to preclinical neurological studies involving autonomic nervous system manipulation. It can be used for direct nerve injection for other nerves in rats and other species, with necessary modifications.

Here we describe a low-cost, rapid, controlled and uniform fixation procedure using 4% paraformaldehyde perfused via the vascular system: through the heart of the rat to obtain the best possible preservation of the brain.

Iris Fixation via External Pentagram Suturing

The present protocol describes how a 10-0 polypropylene suture crosses the anterior surface of the iris, forming a pentagram to prevent both iris and pupil from moving toward the cornea. This can be combined with subsequent keratoplasty to cure bullous keratopathy with extensive anterior synechia of the iris.
