Basal Potassium Stimulation

Basal potassium stimulation is a neuroscience method that uses a controlled increase in extracellular potassium to activate neurons and assess their functional responses under near-baseline experimental conditions. Elevated potassium reduces the transmembrane potassium gradient, depolarizing neuronal membranes and opening voltage-gated calcium channels; the resulting calcium influx promotes neurotransmitter release. Researchers use this approach to examine neuronal excitability, synaptic transmission, and the regulation of chemical signaling in neural tissue or isolated preparations. Comparing stimulated responses with basal measurements can reveal how drugs, disease states, or experimental manipulations alter neurotransmitter dynamics and neuronal function.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.
