Convection-enhanced Delivery

Convection-enhanced delivery (CED) is a drug-administration technique that infuses therapeutic agents directly into tissue, especially the brain, to improve localized treatment of conditions affected by limited drug penetration. A catheter creates a controlled pressure gradient that drives bulk flow through the extracellular space, distributing molecules beyond the infusion site and bypassing much of the blood-brain barrier. In pharmacology, CED supports the study and delivery of chemotherapeutics, biologics, gene-based agents, and neuroactive compounds for brain tumors and neurological disorders. Its effectiveness depends on catheter placement, infusion rate, tissue properties, and drug distribution, making these factors central to therapy design and evaluation.

This video demonstrates a convection-enhanced delivery (CED) technique for antibody delivery into the mouse brain. The skull of an anesthetized mouse is exposed, and a burr hole is created at the target area for antibody delivery. A step catheter containing antibodies is inserted at the target site, and via an infusion pump, a pressure gradient is created to deliver antibodies at a controlled rate.

Convection-enhanced delivery (CED) has been proposed as a treatment option for a wide range of neurological diseases. In order to prepare health care professionals for adoption of CED, accessible training models are needed. We describe the use of agarose gel as such a model of the human brain for testing, research, and training.

Convection-enhanced delivery (CED) is a method enabling effective delivery of therapeutics into the brain by direct perfusion of large tissue volumes. The procedure requires the use of catheters and an optimized injection procedure. This protocol describes a methodology for CED of an antibody into a mouse brain.
