Annular Liquid Pool

An annular liquid pool is a ring-shaped region of liquid occupying the space between two approximately concentric boundaries, making it a useful configuration for studying confined fluid behavior. Its shape and stability result from the balance among hydrostatic pressure, surface tension, viscosity, and, when present, forces produced by rotation or imposed flow. Researchers use annular pools to investigate free-surface deformation, wave motion, fluid instabilities, and heat or mass transfer in curved geometries. These systems provide controlled models for testing fluid-dynamics theories and designing processes involving coatings, rotating equipment, thermal management, and other technologies that rely on thin or confined liquid layers.

This protocol demonstrates the basic experimental configuration for water entry experiments with free-falling spheres. Methods for the alteration of liquid surface with penetrable fabrics, the preparation of chemically non-wetting spheres, and steps for splash visualization and data extraction are discussed.

Detection of Rare Genomic Variants from Pooled Sequencing Using SPLINTER

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

MISSION LentiPlex Pooled shRNA Library Screening in Mammalian Cells

Here we use a human LentiPlex pooled library and traditional sequencing methods to identify gene targets promoting cell survival. We demonstrate how to set up and deconvolute a LentiPlex screen and validate the results.
