High-resolution Tandem Mass Spectrometry for Studying Chemical Constituents of Gynura bicolor DC

BFH의 화학적 조성 식별을 대표 결과를 표시하기 위한 모델로 사용하였다. 용매 추출된 Gynura bicolor DC의 기본 피크 크로마토그램. 는 보충 파일 1-보충 그림 S1-S6에 나타내며, 관찰된 머무름 시간(RT), 성분명, 공식, 질량 대 전하 비율(m/z) 및 질량 오차는 표 S1-S6에 나열되어 있습니다. ZBTK에서는 UPLC-TOF-MS의 분리된 피크에서 35가지 종류의 화합물을 모두 식별했습니다. 보충 파일 1-보충 표 S1에 나타난 바와 같이, 주요 화학물질은 산(퀴닉산, DL-말산, 시트르산, o-쿠마르산, 및 3-O-트랜스-쿠마로일퀸산), 배당체(겐티오피크린, 피크로시드 II, 서프루티코시드 J, 코폴린, 및 보르네올-2-O-β-D-아피오푸라노실-(1→6)-β-D-글루코시드) 및 에스테르(빌로발라이드, 트리메틸시트레이트, 및 트랜스-O-글루코실-메틸-트랜스-신나메이트)를 함유하였다. ZBTK-CT에서는 25종의 추가 화합물이 검출되었고(보충 파일 1-보충 표 S2), 45종의 총 화합물이 관찰되었다. 
클로로포름을 추출제로 사용했을 때 50가지 화합물이 모두 발견되었습니다. 그 중 28종의 추가 화합물에는 코로나산(coronaric acid), 산렝산(sanleng acid), 아그리몰 A(agrimol A) 및 메틸아르테미세이트(methyl artemisate)가 포함되었다(보충 파일 1-보충 표 S3). 석유 에테르를 추출제로 사용했을 때 44 종류의 총 화합물이 관찰되었습니다. notopterol, onjisaponin 및 cnideol B를 포함한 17 종류의 추가 화합물을 발견했습니다 (Supplemental File 1-Supplemental Table S4). 총 65 개의 화합물을 추출제로 사용하여 에틸 아세테이트를 사용하여 용매에서 수득하였다. 앞의 4가지 추출물과 비교하여, 다프네틴, 아세토바닐론 B, 스티그마스테롤-3-O-β-D-글루코시드 등 37종의 추가 화합물이 확인되었다(보충 파일 1-보충 표 S5). 부틸 알코올 추출물에서 우리는 렌교사이드 C, 크로몬, 아투라메텔린 F 및 데아세틸마트리카린-8-O-β-D-글루코피라노시드를 포함한 26종의 추가 화합물을 포함하는 67종의 총 화합물을 발견했습니다(보충 파일 1-보충표 S6). 요약하면, 우리는 6 개의 추출제의 모든 화합물 결과를 기반으로 168 개의 화합물을 발견했습니다.
UPLC-TOF-MS에서 화합물 유형의 절단 경로를 깊이 이해하기 위해 세 가지 화합물을 예로 선택했습니다. 산의 경우, m/z = 337.09222인 3-O-트랜스-쿠마로일퀴닉산은 가수분해 반응을 통해C7H10O5 또는C9H8O2 그룹을 손실하여 각각 m/z = 163.04116 또는 199.05557인 중간체를 형성할 수 있습니다(그림 4D). 에스테르의 경우, m/z = 385.11308인 트랜스-O-글루코실-메틸-트랜스-신나메이트는 다른 부위에서 C-C 절단을 통해 m/z = 153.07485 또는 135.04679의 중간체로 전환될 수 있습니다(그림 4E). 배당체의 경우, m/z = 385.11308인 시트러스 C는 지방족 사슬의 C-C 결합 절단을 통해 m/z = 272.09174의 중간체로 변형된 반면, C7H6O2 그룹의 손실은 m/z = 113.02534의 중간체로 이어졌습니다(그림 4F). 또 다른 경로에서는 벤젠 고리의 COC 결합이 끊어지면서 m/z = 163.11245의 중간체가 형성되고 헥소스 단위가 사라졌습니다.
분자 도킹의 대표적인 결과로 그림 4G,H에서 각각 2D 및 3D 구조를 나타냈습니다. 유전자 온톨로지 농축 분석은 생물학적 과정(BP), 세포 구성 요소(CC) 및 분자 기능(MF; 그림 4I). Kyoto Encyclopedia of Genes and Genomes 경로 주석 분석은 차등적으로 발현된 유전자의 경로 결과를 보여주었으며, 이는 유전자의 기능을 더 잘 이해하는 데 사용할 수 있습니다(그림 4J). 분자 도킹의 상세한 결과는 보충 파일 1-보충 표 S7 및 보충 파일 1-보충 그림 S7-S9에 나타내어져 있습니다.
