Transgenic Plants

Transgenic plants are produced by introducing a foreign gene into a plant&#8217;s genome. These plants can be developed for a variety of reasons, including virus or pest resistance, higher yield, or therapeutic protein production.
To produce a transgenic plant, a gene of interest and its promoter are introduced into the plant protoplast, a plant cell without a cell wall, by incubating both in a medium containing polyethylene glycol.
The recombinant protoplast is cultivated on a sterile medium in the presence of growth regulators and nutrients.
The cells of the protoplast grow indefinitely, producing a mass of undifferentiated cells called a callus. Some cells of the callus, called totipotent cells, give rise to new shoots and roots, which can develop into a whole new plant.
Transgenic plants can also be created using the bacteria Agrobacterium tumefaciens, a gram-negative bacterium found in soil. This bacteria is a pathogen that normally causes disease in plants and contains a Ti -plasmid that can integrate into a plant cell genome.
The Ti plasmid has two regions of interest- T-DNA or transfer DNA region which can be inserted into the host cell genome and the virulence region to aid the transfer of T-DNA into the host cell.
This plasmid is modified by inserting a desired gene into the T-DNA region and a gene for antibiotic resistance to enable selective growth of plasmid-containing organisms. The recombinant plasmid is then inserted into the bacteria by applying an electric pulse to increase cell permeability and enable the uptake of the plasmid into the bacteria.
The Agrobacterium tumefaciens carrying recombinant plasmid is grown in a culture medium containing antibiotics.
Plant parts such as the cotyledonary nodes from a tomato plant are cut and incubated in the medium containing bacteria.
The wounded plant cells at the edge of the stem node release compounds that attract the bacteria. The bacteria infects the plant and releases the recombinant plasmid, which then transfers the foreign gene into the plant cell genome.
Only the transformed cells with recombinant DNA and antibiotic-resistant genes survive in the medium containing antibiotics.
The transformed cells now form a callus and develop into a new transgenic plant carrying a transgene.

Recombinante DNA-technologie, transgenese genaamd, wordt vaak gebruikt om een vreemd gen toe te voegen of een schadelijk gen uit een organisme te verwijderen. Dergelijke genetisch gemodificeerde organismen worden transgene organismen genoemd.

De allereerste transgene plant was een tabaksplant die in 1983 werd ontwikkeld en die resistentie vertoonde tegen het tabaksmozaïekvirus. Sindsdien zijn veel transgene planten ontwikkeld en op de markt gebracht voor het verbeteren van de land-, sier- en tuinbouwwaarde van een gewasplant. Transgene planten kunnen ook worden gebruikt als bioreactor voor de productie van economisch belangrijke metabolieten of eiwitten.

Fysische methoden voor het overbrengen van genen naar planten

Er kunnen veel methoden worden gebruikt, zoals elektroporatie en microprojectielbombardement, om een vreemd gen in een plantenprotoplast over te brengen. Tijdens elektroporatie levert een apparaat hoogspanningspulsen aan de protoplastsuspensie. Dit verhoogt de permeabiliteit van de protoplast waardoor het vreemde DNA de cel kan binnendringen en in het plantengenoom kan integreren.

Tijdens het bombarderen van microprojectielen of de biolistische methode worden bolvormige deeltjes gemaakt van wolfraam of goud (microprojectielen) daarentegen gecoat met het plasmide dat het betreffende gen draagt. De gecoate deeltjes worden vervolgens versneld tot 300 tot 600 m/s met een instrument dat een deeltjeskanon wordt genoemd. Het instrument maakt gebruik van heliumgas onder hoge druk om voortstuwingskracht te leveren die de met plasmide gecoate deeltjes helpt de plantencelwand te penetreren. Eenmaal in de cel integreert het betreffende gen in het plantengenoom. Deze methode kan worden gebruikt om DNA te introduceren in callusculturen, meristeemweefsels, onrijpe embryo's, plantencelsuspensies, enz.

Toepassingen van transgene planten

De recombinant-DNA-technologie wordt veel gebruikt bij de ontwikkeling van plaag- en virusresistente planten. De techniek is ook nuttig bij het wijzigen van het lipiden-, zetmeel- en eiwitgehalte van de zaden.

Gouden rijst is bijvoorbeeld een genetisch gemodificeerde rijstplant met verbeterde voedingswaarde. Deze transgene rijstsoort produceert β-caroteen, een voorloper van vitamine A, waardoor de rijst een gouden kleur krijgt. Het wordt geproduceerd door Agrobacterium-gemedieerde DNA-transformatie, waarbij het recombinante DNA dat drie genen draagt die coderen voor enzymen: fytoeensynthase, fytoeendesaturase en lycopeen β-cyclase, wordt ingevoegd in het genoom van beoogde rijstembryo's. De embryo's ontwikkelen zich vervolgens tot planten die β-caroteen produceren. Gouden rijst is ontwikkeld om vitamine A-tekort in ontwikkelingslanden aan te pakken.

Sommige andere transgene planten, zoals Bt-katoen, zijn gemodificeerd door het inbrengen van een gen dat is geïsoleerd uit de bacterie Bacillus thuringiensis - een grampositieve bacterie die in de bodem wordt aangetroffen. Dergelijke transgene planten produceren kristal- of ‘Cry’-eiwit dat schadelijke insecten doodt, waardoor de plant insectenresistent wordt.

Transgenic plants are crops that have been changed with recombinant DNA technology, also called transgenesis. This method adds a foreign gene or removes a harmful one from an organism. The result is a genetically modified organism. The first transgenic plant was a tobacco plant made in 1983, and it was resistant to tobacco mosaic virus.

Since then, transgenic plants have been developed for many uses in agriculture, horticulture, and ornamental plants. Some are designed to resist pests or viruses. Others are used to improve the lipid, starch, or protein content of seeds. In some cases, transgenic plants can also act as bioreactors and make useful metabolites or proteins.

Foreign genes can be transferred into plant protoplasts by physical methods such as electroporation and microprojectile bombardment. In electroporation, high-voltage pulses make the protoplast membrane more permeable. This allows the foreign DNA to enter the cell and integrate into the plant genome. In microprojectile bombardment, also called the biolistic method, tungsten or gold particles are coated with plasmid DNA and shot into the cells with a particle gun.

The particle gun uses high-pressure helium gas to accelerate the coated microprojectiles to about 300 to 600 m/s. The particles can penetrate the plant cell wall and deliver the gene of interest. This method can be used with callus cultures, meristematic tissues, immature embryos, and plant cell suspensions. Once inside the cell, the introduced DNA can integrate into the plant genome.

A well-known example is golden rice, which has enhanced nutritional value. It produces β-carotene, a precursor of vitamin A, and this gives the rice its golden color. Golden rice was made by Agrobacterium-mediated DNA transformation using recombinant DNA with genes for phytoene synthase, phytoene desaturase, and lycopene β-cyclase. It was developed to help address vitamin A deficiency in developing nations.

Another example is Bt cotton. It contains a gene from Bacillus thuringiensis, a gram-positive soil bacterium. The plant makes Cry protein, which kills harmful insects and makes the crop insect resistant. These examples show how transgenic plants can improve crop value and help solve real agricultural problems.
