A centrosome is the main microtubule organizer of a eukaryotic cell and plays a vital role in the formation of the mitotic spindle, a bipolar arrangement of microtubules that separates sister chromatids. The two spindle poles are formed by two oppositely placed centrosomes.
Prior to the cell cycle, however, most animal cells contain only one centrosome in the cytoplasm. The G1/S-cyclin dependent kinase or Cdk complex&#8212;consisting of cyclin E and Cdk2 in animal cells&#8212;helps trigger centrosome duplication as well as cell cycle entry.&#160;
Each centrosome consists of two centrioles, which are encapsulated by a proteinaceous pericentriolar matrix.
The centrosome duplication starts in the early G1 phase when the separase carries out the disengagement of the two centrioles. A protein belonging to the polo-like protein kinase family, Plk-4, is an important regulator for centriole biogenesis in humans.
Near the onset of S phase, a daughter centriole starts growing at a right angle to the base of each centriole.
During G2, the daughter centrioles typically finish elongating. The four centrioles remain close together in the centrosome until the cell enters mitosis.
At the beginning of mitosis, the centrosome splits in two, which is also known as centrosome disjunction. The two resulting centrosomes each contain a pair of centrioles and the pericentriolar matrix. Both centrosomes nucleate a set of microtubules, primarily at the mother centriole, collectively forming the mitotic spindle.
Centrosome and chromosome life cycle are similar in several aspects. Their duplication occurs only once per cycle and both duplicate through a semiconservative process where a new copy is produced from the pre-existing parent copy.
Also, like chromosomes, one copy of the centrosome is equally distributed to each daughter cell after the division.
Excessive centrosome duplication can impair mitotic spindle assembly and cause chromosomes to segregate improperly, which can cause diseases like cancer.

Het primaire microtubuli-organiserende centrum (MTOC) in dierlijke cellen is het centrosoom. Een centrosoom heeft twee cilindrische centriolen in de kern. Elke centriol bestaat uit negen sets van drie microtubuli die bij elkaar worden gehouden door eiwitten. De centriolen zijn loodrecht op elkaar geplaatst en omgeven door een vormeloze eiwitwolk die de pericentriolaire matrix of pericentriolair materiaal (PCM) wordt genoemd.

Om ervoor te zorgen dat elke dochtercel na de celdeling een centrosoom krijgt, begint de centrosoomduplicatie vroeg in de celcyclus. Centrosoomduplicatie wordt strak gereguleerd door celcycluscontroles – zoals cycline-afhankelijke kinase 2 (Cdk2) – om te voorkomen dat dit meer dan één keer per celcyclus voorkomt. Dus tegen de tijd dat de cel mitose bereikt, heeft deze twee centrosomen.

Centrosoomafwijkingen en defecten in de progressie van de centrosoomcyclus zijn betrokken bij meerdere ziekten, met name kanker. Centrosome duplication is a key part of the animal cell cycle. The centrosome is the main microtubule organizing center, or MTOC, in animal cells. It contains two cylindrical centrioles at its core. Each centriole has nine sets of three microtubules held together by proteins. The two centrioles sit at right angles and are surrounded by the pericentriolar matrix, or PCM, a shapeless protein cloud.

Duplication starts early in the cell cycle so each daughter cell can receive a centrosome after cell division. Cell cycle controls keep this process tightly regulated. Cyclin-dependent kinase 2, or Cdk2, helps prevent duplication from happening more than once in a single cell cycle. By the time the cell reaches mitosis, it has two centrosomes.

Centrosome duplication matches the stages of the cell cycle. In G1, the two centrioles separate. This separation is called centrosome disorientation. During G1 and S, new centrioles called procentrioles begin to form at the base of each older centriole. The procentrioles then elongate through S and G2 until they are the same size as the older centrioles.

The four centrioles stay close together inside the enlarged PCM until mitosis begins. During G2, gamma-tubulin and other PCM proteins build up in the centrosome. This step is called centrosome maturation. Between G2 and M, the centrosomes start to separate. The two mother centrioles disconnect, and microtubule motor proteins move the centrosomes apart.

Problems in centrosome regulation can lead to abnormal numbers of centrosomes and chromosomes. These defects are linked to several diseases, especially cancer. Tumor suppressor proteins and oncogenes are associated with harmful changes in tumor cell centrosomes. Because of this, they are important targets for treatment research.
