Separation of Sister Chromatids

At the end of metaphase, the bi-oriented chromosomes align at the metaphase plate. During this phase, the cohesin protein ring-complexes hold the sister chromatids together at the centromere region and prevent them from being pulled apart.
Metaphase to anaphase progression is triggered by the cyclin-Cdk-induced phosphorylation of a multisubunit, ubiquitin ligase enzyme &#8211; the anaphase-promoting complex, also known as the cyclosome or APC/C. Phosphorylated APC/C binds to a protein, Cdc20, forming an active complex.
The active APC/C complex recognizes an inhibitory protein called securin that is bound to a protease enzyme called separase. Prior to this recognition, securin inhibits the activity of separase.
The active APC/C complex tags securin with the protein ubiquitin, targeting it for proteasomal degradation. Destruction of securin releases separase.
The cyclin-Cdk complex also negatively regulates unbound separase activity through inhibitory phosphorylation.
The active APC/C complex causes the ubiquitination of cyclins, targeting them for proteasomal degradation. The destruction of cyclin removes the enzymatic activity of cyclin-dependent kinases or Cdks.
Inactivation of Cdks allows phosphatase enzymes to dephosphorylate separase and prevents Cdk-mediated inhibitory re-phosphorylation of separase. Dephosphorylation allows the separase enzyme to cleave the cohesin ring-complex that holds sister chromatids together.
Cohesin cleavage results in the absolute loss of sister-chromatid cohesion, marking the transition from metaphase to anaphase. The loss of cohesion permits the sister chromatids to separate and move to opposite poles of the spindle.

Bij het begin van de anafase wordt separase, een proteolytisch enzym, geactiveerd. Het geactiveerde separase splitst de Scc1-subeenheid van de resterende cohesineringen, wat resulteert in een totaal verlies aan cohesie. De zusterchromatiden gaan uit elkaar als er geen kracht is om ze bij elkaar te houden. Het gebrek aan cohesie maakt het mogelijk dat de poolwaartse krachten, die langs de microtubuli spannen, de gescheiden chromatiden naar de spoelpolen trekken.

Bij vroege mitose verbiedt het controlepunt van de spoelassemblage (SAC) dat het anafase-bevorderende complex of cyclosoom (APC/C) eiwitten alomtegenwoordigt, zoals securine en M-fase-cyclinen, waarvan de afbraak noodzakelijk is voor anafase. Alleen wanneer elk chromosoom correct is uitgelijnd op de mitotische spoel, wordt de SAC gedeactiveerd om fosforylering en activering van APC/C-regulerende subeenheden – CDC20 en CDH1 – mogelijk te maken.

De CDC20 wordt gefosforyleerd door Cdk1/cycline B om actief APC/C-CDC20 te vormen. Actief APC/C-CDC20 katalyseert vervolgens de afbraak van securine, cycline A en cycline B om de anafase-overgang te bevorderen. Omdat Cdk-activiteit cycline-afhankelijk is, resulteert de afbraak van cycline B in het verlies van Cdk1-activiteit. Verlies van het Cdk1/cycline B-complex inactiveert APC/C-CDC20 maar activeert een andere regulerende subeenheid, APC/C-CDH1, die de voltooiing van de metafase-naar-anafase-overgang signaleert. De actieve APC/C-CDH1 vergemakkelijkt de mitotische exit en stabiliseert de daaropvolgende G1-fase door de voortijdige accumulatie van mitotische cyclinen te voorkomen.

De omschakeling in de activiteiten van APC/C-CDC20 en APC/C-CDH1 heeft twee belangrijke gevolgen. Ten eerste veroorzaken deze regulerende subeenheden overlappende maar toch verschillende substraatspecificiteiten en bevorderen daardoor een geordende celcyclustransitie. Ten tweede worden CDC20 en CDH1 gereguleerd via verschillende mechanismen. Wanneer APC/C-CDC20 actief is, ondergaat CDH1 remmende fosforylatie door cycline B/Cdk1, waardoor het niet kan binden aan APC/C. Daarentegen wordt de CDC20-activiteit geremd door een mitotisch controlepuntcomplex: een multiproteïne (BUBR1, BUB3, CDC20 en MAD2) geactiveerd door de SAC.

Anaphase starts when sister chromatids are released from one another and pulled toward opposite spindle poles. Before that happens, cohesion along the chromosomal arms is reduced from prophase to metaphase, which helps the sister chromatids resolve. A residual cohesin link still holds them together until the metaphase-to-anaphase transition. This leftover connection helps prevent early separation and reduces the risk of aneuploidy, or the wrong number of chromosomes, in daughter cells.

At the onset of anaphase, separase, a protein-cutting enzyme, becomes active. It cleaves the Scc1 subunit of the remaining cohesin rings. Once that cohesion is lost, the sister chromatids separate without needing a force to pull them apart first. Then the poleward forces along the microtubules can draw the separated chromatids toward the spindle poles.

The spindle assembly checkpoint, or SAC, controls this timing in early mitosis. It blocks the anaphase-promoting complex or cyclosome, called APC/C, from ubiquitinating proteins such as securin and M-phase cyclins until each chromosome is correctly aligned on the mitotic spindle. After alignment, the SAC is turned off. This allows the APC/C regulatory subunits CDC20 and CDH1 to be phosphorylated and activated in sequence.

Phosphorylated CDC20, together with Cdk1/cyclin B, forms active APC/C-CDC20. This complex promotes anaphase by triggering the degradation of securin, cyclin A, and cyclin B. Because Cdk activity depends on cyclins, cyclin B breakdown lowers Cdk1 activity. That loss inactivates APC/C-CDC20 and activates APC/C-CDH1, which signals that the metaphase-to-anaphase transition is complete.

APC/C-CDH1 helps cells exit mitosis and stabilize the next G1 phase by stopping mitotic cyclins from building up too early. The switch between APC/C-CDC20 and APC/C-CDH1 has two important effects. It gives these regulators overlapping but different substrate targets, which helps keep the cell cycle in order. It also reflects different control systems. When APC/C-CDC20 is active, CDH1 is held back by inhibitory phosphorylation from cyclin B/Cdk1. In contrast, CDC20 is blocked by the mitotic checkpoint complex, which includes BUBR1, BUB3, CDC20, and MAD2 and is activated by the SAC.
