Collagen Ii Alpha I

Collagen II alpha I is the alpha-1(II) chain encoded by COL2A1, a major structural component of cartilage and other specialized connective tissues. Three alpha-1(II) chains assemble into a triple-helical type II collagen molecule, which forms fibrils within the extracellular matrix and helps tissues withstand mechanical stress while supporting cell organization. In bioengineering, collagen II alpha I guides the design of cartilage scaffolds, biomaterials, and engineered tissue models that reproduce native matrix properties. Studying its structure, assembly, and interactions also supports research into cartilage development, degeneration, inherited skeletal disorders, and strategies for regenerative medicine.

Simple and reproducible procedures are described for making three structurally distinct collagen assemblies from a common commercially available Type I collagen monomer. Native type, fibrous long spacing or segmental long spacing collagen can be constructed by varying the conditions to which the 300 nm long and 1.4 nm diameter monomer building block is exposed.

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.
