Monocyte Derived Macrophages

Monocyte-derived macrophages are immune cells generated when circulating blood monocytes differentiate into macrophages under tissue or laboratory conditions, providing a useful model of innate immune function. In culture, signals such as macrophage colony-stimulating factor and local inflammatory cues drive changes in cell morphology, gene expression, phagocytosis, and cytokine production. In bioengineering, these cells help evaluate how biomaterials, implants, scaffolds, and engineered tissues interact with the immune system. Their responses can reveal inflammatory or regenerative effects, supporting the design of materials that promote biocompatibility, controlled immune modulation, and improved tissue repair.

Differentiation and Polarization of Monocyte-Derived Cells into Macrophage-Like Cells

2025

In this video, we demonstrate a procedure to induce the differentiation and polarization of peripheral blood-derived monocytes into M1 or M2 macrophages. Incubation with granulocyte-macrophage colony-stimulating factor, lipopolysaccharide, and interferon‐gamma yields pro-inflammatory M1 macrophages, while incubation with macrophage colony-stimulating factor and interleukin-4 induces anti-inflammatory M2 macrophage production.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

A Fully Differentiated Macrophage Infection Model of Monocyte-Derived Cells with Mycobacterium tuberculosis

This video demonstrates a method to study the responses to Mycobacterium tuberculosis infection in human M1 and M2 polarized monocyte-derived macrophages. The use of flow cytometry enables the study of a variety of surface markers associated with infected and uninfected macrophage populations.

Visualization of Caspase Activation in Human Monocyte-Derived Macrophages

This video demonstrates the activation of caspase proteins in human macrophages using bimolecular fluorescence complementation reporter proteins. The pro-domains of caspase-1 are fused with non-fluorescent fragments of Venus proteins, which are recruited to inflammasome complexes. The proximity of inflammasomes induces the refolding and fluorescence of Venus fragments, confirming caspase activation in the macrophages.
