Through Focus Image Acquisition

Through-focus image acquisition is a microscopy method that records a series of images while systematically changing the focal plane, providing information about specimen structure and axial position beyond a single in-focus image. During acquisition, the microscope steps the objective or sample through defined focal positions, producing an image stack in which features change in sharpness, intensity, or appearance with defocus. Computational analysis can use these variations to estimate depth, reconstruct three-dimensional morphology, or characterize optical properties. In bioengineering, the method supports noninvasive imaging of cells, tissues, microstructures, and engineered biological systems when volumetric information is important.

We describe a detailed protocol using high-resolution episcopic microscopy to acquire three-dimensional (3D) images of mouse embryos. This improved protocol utilizes a modified tissue preparation method to enhance penetration of the fluorescent dye, thereby permitting morphometric analysis of both small and large-sized specimens.

This article describes real-time monitoring of HIFU ablation in canine liver with high frame rate ultrasound imaging using diverging and plane wave imaging. Harmonic Motion Imaging for Focused Ultrasound is used to image the decrease of acoustic radiation force induced displacement in the ablated region.
