Alpha-1 Antitrypsin Deficiency

Alpha-1 antitrypsin deficiency is an inherited disorder in which mutations in SERPINA1 reduce production or alter folding of alpha-1-antitrypsin, a protein that protects lung tissue from excessive protease activity. In common severe variants, misfolded protein accumulates in liver cells while too little reaches the bloodstream, allowing neutrophil elastase to degrade alveolar connective tissue and potentially cause emphysema; liver injury can also result. Genetic testing, serum protein measurement, and pulmonary and hepatic assessment help diagnose and monitor the condition, while augmentation therapy, smoking avoidance, and targeted management can reduce complications.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.

The ruthenium-catalyzed olefination of electron-deficient alkenes with allyl acetate is described here. By using aminocarbonyl as a directing group, this external oxidant-free protocol has high efficiency and good stereo- and regioselectivity, opening a novel synthetic route to (Z,E)-butadiene skeletons.
