Alpha-sma-gfp Mice

Alpha-SMA-GFP mice are transgenic reporter animals that visualize cells expressing alpha smooth muscle actin, a cytoskeletal protein associated with smooth muscle cells and activated myofibroblasts. In this model, regulatory sequences from the alpha-smooth muscle actin gene drive production of green fluorescent protein, causing relevant cells to emit fluorescence that can be detected by microscopy or used for cell isolation. Researchers use Alpha-SMA-GFP mice to study smooth muscle development, vascular biology, wound repair, and tissue fibrosis, including how contractile or myofibroblast-like cells arise, migrate, and contribute to tissue remodeling. The model supports spatial and temporal analysis of these processes in living tissues and disease models.

Peripheral injection of alpha-synuclein fibrils into the peritoneum or tongue of Tg(M83+/-:Gfap-luc+/-) mice, which express human alpha-synuclein with the familial A53T mutation and firefly luciferase, can induce neuropathology, including neuroinflammation, in their central nervous system.

Induction of Neuroinflammation in Mice via Injection of Alpha-Synuclein Fibrils

2025

This video demonstrates the injection of alpha-synuclein fibrils into a mouse to induce neuroinflammation. The fibrils are injected intraperitoneally into a transgenic mouse expressing mutated alpha-synuclein proteins. Upon reaching the central nervous system, the fibrils enter neurons and induce aggregation of the mutated proteins. The aggregates spread to neighboring neurons, microglia, and astrocytes. Recognition of the aggregates triggers microglia to induce neuroinflammation, damaging the...

Two Techniques to Create Hypoparathyroid Mice: Parathyroidectomy Using GFP Glands and Diphtheria-Toxin-Mediated Parathyroid Ablation

Mice with acquired hypoparathyroidism would be useful for studying novel drug therapies for hypoparathyroidism. Two procedures to create such mice are demonstrated. The GFP-PTX mouse is generated by surgical parathyroidectomy guided by green fluorescing parathyroid glands. A second, non-surgical approach is based on parathyroid-specific expression of the diphtheria toxin receptor.
