Axon Glial Interactions

Axon-glial interactions are the reciprocal communications between neuronal axons and glial cells that regulate nervous-system development, maintenance, and signaling. In the central and peripheral nervous systems, oligodendrocytes and Schwann cells ensheath axons with myelin, while glia also provide metabolic support, organize ion homeostasis, and influence nodes of Ranvier where action potentials are regenerated. These interactions shape conduction speed, axonal survival, and responses to injury or inflammation. Studying them helps explain neurological disorders involving demyelination and guides research into nerve repair, neuroprotective therapies, and engineered neural tissues.

This study describes the procedures of setting up a novel neuronal axon and (astro)glia co-culture platform. In this co-culture system, manipulation of direct interaction between a single axon (and single glial cell) becomes feasible, allowing mechanistic analysis of the mutual neuron to glial signaling.

Although the peripheral nervous system (PNS) is capable of significant repair after injury, little is known about the cellular and molecular mechanisms that govern this phenomenon. Using live, transgenic zebrafish and a reproducible nerve transection assay, we can study dynamic glial cell behaviors during nerve degeneration and regeneration.

We described structural features of the Glia-neuromuscular synapses in a novel Inside-out tissue preparation of live fly larvae using fluorescent dyes with confocal microscopy. We labeled live neuron terminals with fluorescent primary antibodies to HRP, and also visualized the perisynaptic space with fluorescent Dextrans.
