Cell Lysate Preparation

Cell lysate preparation is the process of breaking open cells to release intracellular proteins, nucleic acids, metabolites, and organelles for laboratory analysis. It typically combines mechanical disruption, chemical detergents, enzymatic digestion, or osmotic shock with a suitable buffer, while controlling temperature and protease activity to preserve target molecules. The resulting lysate supports methods such as Western blotting, enzyme assays, immunoprecipitation, mass spectrometry, and nucleic acid analysis. In biology, carefully selected lysis conditions determine extraction efficiency, sample integrity, and reproducibility, making preparation a critical foundation for studying cellular composition, signaling, gene expression, and disease-related molecular changes.
