Cell Resuspension

Cell resuspension is the process of evenly redistributing pelleted cells in a liquid medium after centrifugation or settling, a routine step that supports accurate handling and analysis in biology. It works by adding an appropriate buffer or culture medium and using controlled pipetting or gentle agitation to separate cells while limiting shear stress, aggregation, and loss of viability. The resulting suspension can be adjusted for cell concentration and used in cell counting, culture, staining, flow cytometry, nucleic acid or protein extraction, and other assays. Consistent resuspension improves sample uniformity, measurement reliability, and experimental reproducibility.

Quantifying Bulk Autophagic Sequestration Activity in Mammalian Cells

2025

The video demonstrates an assay for assessing bulk autophagic sequestration activity in mammalian cells. It utilizes electroporation to release cytosolic proteins, including lactate dehydrogenase (LDH). The sequestered LDH and total cellular LDH fractions are determined to assess LDH sequestration, providing insights into the overall bulk sequestration activity.

Differentiation of Induced Pluripotent Stem Cells into Neural Progenitor Cells

2025

The video demonstrates a technique for differentiating human induced pluripotent stem cells (hiPSCs) into neural progenitor cells (NPCs). Initially, the hiPSCs are cultured on a feeder layer of mouse embryonic fibroblasts (MEFs). Subsequently, the cells are detached and transferred to a biopolymer-coated plate to allow the MEFs to adhere to and separate from the hiPSCs. The suspended hiPSCs are then transferred to a V-bottom plate and centrifuged to form aggregates. Finally, the addition of a...

Isolation of IPE and RPE cells: A Technique to Obtain Pigmented Epithelial Cells from Isolated Porcine Eye
