Embryo Rehydration

Embryo rehydration is the controlled restoration of water balance in embryos after dehydration or cryopreservation, making it important for preserving cellular integrity and viability. The process typically moves embryos through solutions with carefully controlled osmolarity, allowing water to re-enter cells while limiting abrupt volume changes and osmotic injury. In assisted reproduction, rehydration follows vitrification and warming, supporting recovery of embryo structure before culture or transfer. In developmental biology and reproductive research, standardized rehydration helps assess embryo viability and improves post-preservation handling, with outcomes influenced by developmental stage, solution composition, and exposure conditions.

We present a protocol to rapidly form giant polymer vesicles (pGVs). Briefly, polymer solutions are dehydrated on dried agarose films adhered to coverslips. Rehydration of the polymer films results in rapid formation of pGVs. This method greatly advances the preparation of synthetic giant vesicles for direct applications in biomimetic studies.

By combining methods for RNA whole mount in situ hybridization and histology, gene expression can be linked with cell fate decisions in the developing embryo. These methods have been adapted to marine elasmobranchs and facilitate the use of these animals as model organisms for biomedical, toxicology and comparative studies.
