Insect Cell Culture

Insect cell culture is the controlled growth and maintenance of insect-derived cells outside the organism, providing a versatile system for studying cellular biology and producing biological materials. Cells are propagated in nutrient-rich culture media under aseptic conditions, with temperature, pH, oxygen, and cell density adjusted to support survival and proliferation; they may grow as adherent layers or suspended populations. In biology research, insect cell cultures support studies of gene expression, virus replication, host-pathogen interactions, and protein production, particularly through baculovirus-based expression systems. These cultures offer scalable, experimentally tractable models for investigating cellular processes and developing vaccines, enzymes, and other recombinant products.

Transient Expression and Cellular Localization of Recombinant Proteins in Cultured Insect Cells

Nonlytic insect cell expression systems are underutilized for production, cellular trafficking/localization, and recombinant protein functional analysis. Here, we describe methods to generate expression vectors and subsequent transient protein expression in commercially available lepidopteran cell lines. The co-localization of Bemisia tabaci aquaporins with subcellular fluorescent marker proteins is also presented.

In this video, we demonstrate the cytopathic effect assay to determine the tissue culture infective viral dose of Drosophila C virus on Drosophila S2* cells. A range of viral concentrations are used to infect the cultured cells. The proportion of cells exhibiting structural changes is determined, and this data is used to calculate the tissue culture infective dose.
