Sonication Treatment

Sonication treatment is a laboratory technique that uses high-frequency sound waves to disrupt, disperse, or mix biological materials, making it valuable for sample preparation and molecular biology. An ultrasonic probe or bath transfers acoustic energy into a liquid, producing rapid pressure changes and cavitation bubbles whose collapse generates localized shear forces that break cell membranes, detach tissue components, or reduce particle aggregates. Researchers use sonication to lyse cells, release proteins, DNA, and organelles, homogenize suspensions, and prepare samples for downstream analysis. Treatment conditions, including power, duration, temperature, and sample volume, must be controlled to limit heating and protect sensitive biomolecules.

Immunostaining is an effective technique for visualizing specific cell types and proteins within tissues. By utilizing sonication, the protocol described here alleviates the need to dissect Drosophila melanogaster tissues from late-stage embryos and larvae before immunostaining. We provide an efficient methodology for the immunostaining of formaldehyde-fixed whole mount larvae.
