Substrate Feeding Strategy

Substrate feeding strategy is a biological and bioprocess approach for controlling how nutrients or other growth substrates are supplied to cells or microorganisms over time. Rather than providing the entire nutrient supply at once, the strategy adds substrate at a defined rate or in response to measurements such as concentration, growth, or metabolic activity, helping balance nutrient availability with cellular demand. In biology, controlled feeding can reduce substrate depletion, limit inhibitory accumulation, and support sustained growth or product formation. It is therefore important in cell culture, microbial fermentation, bioreactor operation, and studies of metabolism, where precise nutrient control improves reproducibility and process performance.

We demonstrate how to use the RNAi feeding technique to knock down target genes and score body size phenotype in C. elegans. This method could be used for a large scale screen to identify potential genetic components of interest, such as those involved in body size regulation by DBL-1/TGF-β signaling.

The feeding circuit in Drosophila melanogaster larvae serves a simple yet powerful model that allows changes in feeding rate to be correlated with alterations in the stomatogastric neural circuitry. This circuit is composed of central serotonergic neurons that send projections to the mouth hooks as well as the foregut.

Substrate Generation for Endonucleases of CRISPR/Cas Systems

CRISPR/Cas systems mediate adaptive immunity in Bacteria and Archaea. Many Cas proteins are proposed to act as endoribonucleases acting on crRNA precursors of varying length. Here we illustrate three different approaches to generate pre-crRNA substrates for the biochemical analysis of Cas endonuclease activity.
