Basement Matrix

Basement matrix is a specialized extracellular matrix that supports epithelial and endothelial tissues and provides structural and biochemical cues to nearby cells. It is formed mainly from networks of laminins and type IV collagen, linked by proteoglycans and other matrix proteins; integrin-mediated adhesion to these components regulates cell polarity, survival, migration, and differentiation. In cancer research, basement matrix models help examine how tumor cells interact with their microenvironment, cross tissue barriers, and initiate invasion or metastasis. These systems also support three-dimensional culture and drug testing, providing more physiologically relevant measurements than many two-dimensional assays.

The extracellular matrix undergoes substantial remodeling during wound healing, inflammation and tumorigenesis. We present a novel intravital immunofluorescence microscopy approach to visualize the dynamics of fibrillar as well as mesh-like matrix components with high spatial and temporal resolution using epifluorescence or two-photon microscopy.
