Epithelial Regeneration

Epithelial regeneration is the biological process that restores epithelial tissues after injury by replacing damaged or lost cells and reestablishing a protective barrier. It depends on coordinated activation of resident stem and progenitor cells, cell proliferation, migration across the wound, and differentiation, with tissue signals regulating polarity, adhesion, and repair. In cancer research, studying these responses helps clarify how normal wound-healing programs become misregulated, enabling uncontrolled growth, invasion, or treatment resistance. Experimental models of epithelial repair therefore support investigation of tumor initiation and progression while providing frameworks for testing therapies that target regenerative pathways without disrupting essential tissue maintenance.

A System for Culturing Iris Pigment Epithelial Cells to Study Lens Regeneration in Newt

In newt, the lens regenerates always from the dorsal iris by transdifferentiation of the iris pigmented epithelial cells (IPEs). Here we describe a procedure to culture dorsal and ventral newt IPE cells and their implantation to the newt eye. The implanted cells are then studied by tissue sectioning and immunohistochemistry.

Acute kidney injury (AKI) in humans is a common clinical problem caused by damage to the epithelial cells that comprise kidney nephrons, and AKI is associated with high mortality rates of 50-70%1. Following epithelial cell destruction, nephrons have a limited ability to regenerate, though the mechanisms and limitations that guide this phenomenon remain poorly understood. In this video article, we describe our technique for targeted laser ablation of kidney nephron cells in the zebrafish...

The gastrointestinal tract is one of the most sensitive organs to injury upon radiotherapeutic cancer treatments. It is simultaneously an organ system with one of the highest regenerative capacities following such insults. The presented protocol describes an efficient method to study the regenerative capacity of the intestinal epithelium.

An Introduction to Aging and Regeneration

2023

Tissues are maintained through a balance of cellular aging and regeneration. Aging refers to the gradual loss of cellular function, and regeneration is the repair of damaged tissue generally mediated by preexisting adult or somatic stem cells. Scientists are interested in understanding the biological mechanisms behind these two complex processes. By doing so, researchers may be able to use somatic stem cells to treat degenerative diseases and develop therapies that could delay the effects of...
