Erk Expression

ERK expression refers to the production and abundance of extracellular signal-regulated kinase proteins, key components of the mitogen-activated protein kinase (MAPK) signaling network that regulates cell behavior. In cells, growth-factor signals activate receptor-linked RAS–RAF–MEK pathways, while gene transcription and protein turnover determine ERK levels; activated MEK then phosphorylates ERK, enabling it to regulate nuclear and cytoplasmic targets. In cancer research, measuring ERK expression alongside ERK phosphorylation helps distinguish changes in pathway abundance from changes in signaling activity, supporting studies of tumor growth, therapeutic response, resistance, and biomarker development. These analyses can clarify how dysregulated MAPK signaling contributes to malignant progression.

Spatial and Temporal Analysis of Active ERK in the C. elegans Germline

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.

RASopathies are multisystem genetic syndromes caused by RAS-MAPK pathway hyperactivation. Potentially pathogenic variants awaiting validation emerge continuously while poor preclinical evidence limits therapy. Here, we describe our in vivo protocol to test and cross-validate RASopathy-associated ERK activation levels and its pharmacological modulation during embryogenesis by live FRET imaging in Teen-reporter zebrafish.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
