Murine Model Technique

Murine model techniques use mice to reproduce key features of human cancer, providing an experimentally controlled system for studying tumor biology and treatment response. Researchers generate tumors through transplantation of cancer cells, patient-derived xenografts, or targeted genetic alterations, then track tumor growth, metastasis, immune interactions, and responses to therapy under defined conditions. These models help connect molecular mechanisms with disease progression, evaluate candidate drugs and combination treatments, and assess how the tumor microenvironment influences outcomes. Their limitations, including species differences and variable clinical predictiveness, also guide the development of more representative preclinical models.

This video demonstrates a technique to generate a chemical-induced murine model of colitis — an inflammatory bowel disease. The mouse is allowed to consume a dextran sodium sulfate (DSS) solution for an adequate duration. Upon consumption, DSS damages the mucus and epithelial layer barrier in the colon, allowing gut microbes to enter the underlying tissue — where they are sensed by the resident immune cells to mediate an exaggerated immune response, resulting in inflammation.

The murine cervical heart transplantation model is well suited for immunological as well as ischemia reperfusion injury studies. We modified the procedure using a non-suture cuff technique and performed more than 1,000 successful transplants with this approach. Herein, we provide additional details of this technique to supplement the video.
