Tumor Necrosis Evaluation

Tumor necrosis evaluation is the assessment of dead or dying regions within a tumor, a key measure of tissue injury, disease progression, and treatment response in cancer research. Necrosis commonly develops when rapid tumor growth outpaces its blood supply, causing hypoxia, nutrient depletion, cellular energy failure, and loss of membrane integrity. Researchers evaluate necrotic areas using histopathology, imaging, or molecular markers to distinguish viable tumor from damaged tissue. These measurements help characterize tumor aggressiveness, assess how effectively therapies damage cancer cells, and support studies of tumor biology, prognosis, and treatment development.

Evaluation of Tumor-infiltrating Leukocyte Subsets in a Subcutaneous Tumor Model

This protocol describes a method for the detailed evaluation of leukocyte subsets within the tumor microenvironment in a mouse tumor model. Chemerin-expressing B16 melanoma cells were implanted subcutaneously into syngeneic mice. Cells from the tumor microenvironment were then stained and analyzed by flow cytometry, allowing for detailed leukocyte subset analyses.

This protocol describes a detailed workflow for the generation and ex vivo characterization of oncolytic viruses for expression of immunomodulators, using measles viruses encoding bispecific T cell engagers as an example.
